Scientist Mass Spectrometry (Lead Discovery & Characterization)
What you will do
Let’s do this. Let’s change the world. Amgen is a biopharmaceutical leader founded on discovering, developing, manufacturing, and delivering innovative human therapeutics. Amgen's Lead Discovery and Characterization laboratory within Therapeutic Discovery is seeking a highly motivated small molecule Mass Spectrometry expert at Amgen headquarters in Thousand Oaks, CA.
Responsibilities:
The Scientist Mass Spectrometry will be responsible primarily for the development of label-free technologies to monitor and quantify protein-ligand interactions, including screening and characterizing small molecule ligands by Mass Spectrometry. Additional responsibilities may include the development and execution of mass spectrometric-based characterization of protein-small molecule adducts (e.g. native MS or affinity selection MS) as part of our efforts toward the discovery of new drugs.
The successful candidate will work closely with biology and chemistry teams to develop new assays and strategies, analyze results, and present and discuss high quality data in a timely manner and should also contribute to skills development and mentoring for colleagues in a highly multi-disciplinary environment.
What we expect of you
We are all different, yet we all use our unique contributions to serve patients. The professional we seek has these qualifications.
Basic Qualifications:
Doctoral degree
Or
Master’s degree and 2 years of scientific experience
Or
Bachelor’s degree and 4 years of scientific experience
Preferred Qualifications:
- Doctoral degree with 2 years of mass spectrometry experience and a background in quantitative Biochemistry, Cell and/or Molecular Biology, or closely related field.
- Deep knowledge of small molecule MS techniques, especially as applied to monitoring of enzyme reactions for quantitative analysis of protein-ligand interactions and hit identification by ASMS.
- Experience with the operation, troubleshooting, and maintenance of Q-TOF, QQQ, and/or MALDI mass spectrometers, including the use of LC, SPE, or SEC separation techniques.
- Experience in the development of LC-MS, LC-MS/MS, or SPE-MS methods for affinity selection-based MS assays, MS-detected enzyme assays, or to characterize protein-small molecule adducts by intact protein MS.
- Familiarity with Agilent, SCIEX, Bruker instrumentation and software, e.g. Agilent MassHunter and SCIEX MultiQuant software.
- Experience in the development of SPE-MS or MALDI-TOF methods for MS-detected enzyme assays, or to characterize protein-small molecule adducts.
- Fluency in data analysis using GraphPad PRISM, Excel and vendor software. Experience with high-throughput data analysis packages (e.g. Genedata Screener, Expressionist) is an advantage.
- Familiarity with the principles of high-throughput screening (HTS) and experience with microplate readers and automated liquid handling systems Agilent Bravo, Echo, Tecan).
- An understanding of mechanistic enzymology and biophysical assays (SPR, ITC, NMR) for characterization of protein-ligand binding.
- Demonstrated ability to work both independently and as a member of a multidisciplinary team.
- Excellent written and oral communication skills for the timely documentation, presentation, and discussion of science-based results and strategy in a dynamic, multi-disciplinary, team-based environment.
